猪水泡病病毒非结构蛋白编码区的克隆与序列分析

摘要: 以猪水泡病病毒(Swine vesicular disease virus,SVDV)HK’70为材料,用特异性引物扩增出非结构蛋白编码区的4个重叠目的片段,连接于pMD18T载体,转化JM109感受态细胞。经对重组质粒双酶切、PCR鉴定、序列分析表明,HK’70非结构蛋白编码区核苷酸长4002nt,氨基酸长1334aa;与J1’73、H/3’76、SVDV(Seechrn等测株)、NET/1/92的核苷酸同源性分别为98.6%、98.3%、98.3%、91.2%,推导氨基酸序列的同源性分别为98.9%、99.1%、99.2%和97.2%。

Cloning and analysis of nucleotide sequences encoding nonstructure 
protein of swine vesicular disease virus

Abstract: Four overlapping fragments encoding nonstructure protein of swine vesicular disease virus, which had been amplified from SVDV Hk’70 by 4 pairs of primers, respectively, were ligated with pMD18T and transformed into Escherichia coli competent cell JM109, respectively. The recombinant plasmids were identified by restriction enzyme and PCR. Homologyanalysis showed that nonstructure protein sequences of SVDV HK’70 shared 98.6%,98.3%,98.3% and 91.2% homology with that of SVDV J1 73,SVDV H/3’76,SVDV(Seechurn) and SVDV NET/1/92, respectively,and the deduced amino acid sequences from nonstructure protein sequences of SVDV HK’70shared 98.9%,99.1%, 99.2% and 97.2% homology with that from nonstructure protein sequenceof SVDVJ1’73,SVDV H/3’76,SVDV(Seechurn) and SVDV NET/1/92, respectively.
