冬虫夏草菌丝水提液对SP2/0细胞凋亡的影响

在冬虫夏草菌丝水提液诱导下，采用Hoechst33342荧光染色、MTT、琼脂糖凝胶电泳和流式细胞仪等方法，观察和分析了SP2/0细胞的凋亡情况。结果显示，冬虫夏草菌丝水提液可抑制SP2/0细胞增殖，在荧光显微镜下，凋亡细胞呈亮绿色，DNA琼脂糖电泳可见典型的“阶梯状”条带，Annexin-V-FITC和碘化丙啶(PI)双染后流式细胞仪分析显示，细胞凋亡数量明显增多。表明，冬虫夏草菌丝水提液对SP2/0细胞增殖的抑制作用可能与诱导其凋亡有关。 |

Cordyceps sinensis on apoptosis of SP2/0 cells |

Abstract: MTT assay, agarose gel electrophoresis, flow cytometric analysis and Hoechst33342 staining were performed to study apoptosis and its mechanism. After SP2/0 cells were treated with water extract of mycelium, growth of SP2/0 cells was inhibited and SP2/0 cells appeared lighter green in fluorescence microscope. A typical DNA ladder was detected by agarose gel electrophoresis of DNA extracted from SP2/0 cells. Flow cytometric analysis of SP2/0 cells by Annexin-V-Fluos and propidium iodide double staining demonstrated that quantity of apoptosis cell increased with water extract induction. The water extract of mycelium induced apoptosis in SP2/0 cells, indicating that growth of SP2/0 cells was inhibited. The mechanism of anti-tumor activity of water extract of mycelium may be related to induction of apoptosis in tumor cells. |
