鸡IL-2和IL-15基因真核表达质粒对禽流感
疫苗的免疫增强作用

摘要:  
根据GenBank中的鸡IL-2和IL-15序列设计了2对引物,采用RT-PCR法从山地乌骨鸡外周血淋巴细胞RNA中克隆了IL-2和IL-15基因,与pcDNA3.1连接构建了真核表达质粒pDNAIL-2和pDNAIL-15,研究了其对禽流感灭活疫苗的免疫增强作用。结果显示,免疫后第10d,pDNAIL-15+AI灭活疫苗组和pDNAIL-2+AI灭活疫苗组HI抗体水平显著高于AI灭活疫苗组(P<0.05)。免疫后第10d,pDNAIL-15+AI灭活疫苗组CD4+淋巴细胞百分含量显著高于pDNAIL-2+AI灭活疫苗和AI灭活疫苗组(P<0.05);免疫后第23d,pDNAIL-2+AI灭活疫苗组CD4+淋巴细胞百分率显著高于pDNAIL-15+AI灭活疫苗和AI灭活疫苗组(P<0.05)。证实pDNAIL-2和pDNAIL-15能提高禽流感灭活疫苗的免疫效果。

Immunoenhancement of eukaryotic expression plasmids with chicken
IL-2 or IL-15 genes on H5 subtype avian influenza vaccine

Abstract: Two pairs of specific primers for chicken interleukin-2(CHIL-2) and interleukin-15(CHIL-15) genes were designed according to the published gene sequences of CHIL-2 and CHIL-15. The eukar-yotic expression plasmids with CHIL-2 gene or CHIL-15 gene, named pDNAIL-2 and pDNAIL-15, were constructed by subcloning CHIL-2 gene or CHIL-15 gene into pcDNA3.1(+), respectively. The immunoenhancement of pDNAIL-2 and pDNAIL-15 on H5 subtype avian influenza vaccine was observed by detection of HI antibody titers and percentages of CD4+ and CD8+. In results, on day 10 post-inoculation(PI), the HI antibody titers in the pDNAIL-15+AI vaccine group and the pDNAIL-2+AI vaccine group were significantly higher than those in the AI vaccine group(P<0.05). The percentage of CD4+ in the pDNAIL-15+AI vaccine group was significantly higher than that in the pDNAIL-2+AI vaccine group and the AI vaccine group on day 10 PI(P<0.05). The percentage of CD4+ in the pDNAIL-2+AI vaccine group was significantly higher than that in the pDNAIL-15+AI vaccine group and the AI vaccine on day 23 PI(P<0.05).These results indicated that pDNAIL-2 and pDNAIL-15 have adjuvant effects on the AI vaccine.
