江苏省猪高热病病例主要继发感染病毒的
多重PCR检测

摘要:  
应用建立的2个多重PCR方法,对2005~2006年收集于江苏省的211份猪高热病疑似病料进行了检测。结果,PRRSV阳性率为51.2%,PCV2阳性率为44.1%,CSFV阳性率为10.4%,PPV阳性率为3.9%,PRV阳性率为2.4%。2005年病料中PCV2阳性率为21.2%,2006年为73.2%; 2005年PRRSV阳性率为38.1%,2006年为67.7%。2006年猪高热病疫情比2005年严重,PCV2和PRRSV可能在其中发挥了协同致病作用。对10个PRRSV分离株ORF5基因和Nsp2基因进行的测序表明,2006年分离株Nsp2基因中有连续87个碱基的缺失。

Multiplex PCR detection of main subsequent viruses in cases of 
high swine fever in Jiangsu Province from 2005 to 2006

Abstract: Two multiplex PCR assays were developed and their effectiveness was subsequently evaluated to simultaneously detect multiple-virus infection in swine.211 samples from the cases with high swine fever collected from Jiangsu Province were detected for the five viruses,including porcine reproductive and respiratory syndrome virus(PRRSV),porcine circovirus type 2(PCV2),classical swine fever virus(CSFV),porcine parvovirus(PPV) and pseudorabies virus(PRV),using the developed two multiplex PCRs.In result,the positive rates were 51.2%,44.1%,10.4%,3.9% and 2.4% for PRRSV,PCV2,CSFV,PPV and PRV,respectively.The positive rate of PCV2 was 21.2% in 2005,and it increased to 73.2% in 2006.The positive rate of PRRSV was 38.1% in 2005,and it reached 67.7% in 2006.The result showed that the po-sitive rates of PCV2 and PRRSV rose up obviously during the two years,while the positive rates of PPV,PRV and CSFV almost remained the same,and PRRSV and PCV2 infections seemed to become epidemic in Jiangsu Province.The open reading frame 5(ORF5) genes of ten PRRSV isolates and partial Nsp2 genes of six isolates collected in 2005 and 2006 were sequenced,and the results showed that there was a continuous deletion of 87 bases in the Nsp2 gene of PRRSVs isolated in 2006 compared with those of the other PRRSVs isolated before.
