铅镉联合对体外培养大鼠肾小管上皮细胞存活
及凋亡的影响

采用机械筛网结合酶消化法建立了大鼠原代肾小管上皮细胞培养模型,在传代细胞增殖活性最强时间段进行铅、镉单独染毒或联合染毒。通过CCK-8还原法和流式细胞仪检测不同时间铅、镉对细胞存活率和凋亡率的影响。结果显示,铅、镉单独染毒高剂量组从第6h开始、低剂量组从第12h开始,其细胞存活率显著低于对照组(P<0.05);铅、镉联合染毒组从第3h开始,细胞存活率显著或极显著(P<0.05或P<0.01)低于对照组,且存活率的降低幅度与剂量呈正相关;染毒后第12h,各组的凋亡率均极显著高于对照组(P<0.01),铅、镉联合染毒组的细胞凋亡率显著高于各相关单独染毒组(P<0.05),呈明显的剂量-效应关系。 |

and apoptosis of rat’s renal tubular epithelial cells cultured in vitro |

Abstract:The primary renal tubular epithelial cells(RTECs) of rats were cultured by mechanical grinding,filtering and chemical digestive methods.Subcultured cells were used to perform the design when they were in the highest increasing activity.Effects of different doses of lead and cadmium on the survival rates and apoptotic rates at different time points were detected by using the CCK-8 reduction method in the Microplate Reader and the double staining method in the flow cytometer.The cell survival rates in the single lead and in the cadmium groups were significantly lower than those in control groups since these cells were treated for six hours in the high-dose groups and for 12 hours in the low-dose groups,respectively(P<0.05).The cell survival rates in the combined groups were significantly lower than those in the control groups after exposure for 3h(P<0.05 or P<0.01),and the relationship between the degree of decrease in the cell survival rate and dose was in positive correlation.The apoptotic rates in all the treatment groups were very significantly lower than that in control group after exposure for 12h(P<0.01).The apoptotic rates in the combined groups were significantly higher than those in the related single-exposed groups(P<0.05),which was dose-dependent in the process. |
