鸭疫里默氏杆菌培养滤液对鸭胚成纤维细胞
形态的影响

摘要:  
为探索鸭疫里默氏杆菌的致病机制,研究了鸭疫里默氏杆菌培养滤液对鸭胚成纤维细胞形态的影响。结果显示,鸭疫里默氏杆菌AF株的培养滤液能使鸭胚成纤维细胞形态发生变化,表现为细胞膜和细胞质逐渐丢失、核浓缩等;90%(27/30)的鸭疫里默氏杆菌临床分离菌株能液化明胶,具有致病性,培养滤液具有改变鸭胚成纤维细胞形态的生物学活性;10%(3/30)的菌株不液化明胶,无致病性,培养滤液无改变鸭胚成纤维细胞形态的生物学活性;经硫酸铵盐析、阴离子交换、疏水和凝胶过滤层析,从AF株培养滤液中分离获得分子质量约为55ku、能水解明胶及改变细胞形态的活性蛋白质。证实,降解明胶的蛋白水解酶是鸭疫里默氏杆菌培养滤液影响鸭胚成纤维细胞形态的活性物质,可能是细菌的毒力因子之一。

Effect of Riemerella anatipestifer culture filtrate 
on morphology of duck embryo fibroblast

Abstract:  In order to elucidate the pathogenic mechanism of Riemerella anatipestifer(RA),morpho-logy of duck embryo fibroblast(DEF) treated with culture filtrate of RA were observed.DEF treated with the culture filtrate from RA AF strain resulted in striking morphologic changes including losses of cell membrane and cytoplasm,and nucleus being condensed.Most of the clinical strains 90%(27/30) had the capability of hydrolyzing gelatin,causing disease and altering the morphology of DEF,while the rest 10%(3/30) were on the contrary.After treatment by ammonium sulfate precipitation,anion exchange chromatography,hydrophobic chromatography and gel chromatography to the culture filtrate of RA,a protein of approximately 55ku in molecular mass had the above referred biological activity.The results showed that the proteinase with hydrolyzing gelatin activity was relevant to the morphologic changes and it may represent one of bacterial virulence factors.
